When in virtual desktop, go to Steam. When you start AMS2, you should get the popup asking how to open the game. In 2d desktop, in SteamVR or in OculusVR. If you don't see that, you might have set the first option as 'always open in this' and you need to right click, go to properties and set it to always ask.

Start steamvr from the virtual desktop window inside Pico4 not on your computer. Start the game from the steamvr dashboard or via steamvr home.
Was testing a p4 and it works both with steamvr and VD I found convenient to use the GAMES tab on VD dashboard , maybe you should try this way
